Существенным дополнением являются данные об изменении с возрастом содержания агалакто- зилированных форм Fc фрагмента IgG в сыворотке крови человека. Известно, что количество G0 форм увеличивается, а доля сиалированных форм уменьшается. Интересно, что вызванное беременностью галактозилирование и сиалилирова- ние IgG улучшает течение ревматоидного артрита. Также наблюдается снижение галактозы при системных васкулитах, таких как гранулематоз Вегенера и микроскопический полиангиит. Терминальное сиалилирование IgG-гликана можно рассматривать как защитную реакцию при аутоиммунных заболеваниях. Таким образом, введение IgG с сиалированным гликаном в организм оказывает мощное противовоспалительное действие, которое можно использовать в качестве основы для создания гликомодифицированных терапевтических моноклональных антител.

Заключение

Активное применение лекарственных средств на основе иммуноглобулинов и их производных ставит перед специалистами всех уровней новые задачи. Понимание механизма, лежащего в основе терапевтических эффектов, невозможно без изучения фундаментальных принципов функционального воздействия данных молекул на тот или иной физиологический аспект организма. Рассмотренная нами по- странсляционная модификация иммуноглобулинов, а именно различные варианты гликози- лирования, открывает широчайшие возможности как в диагностике иммунологических нарушений, так и в их терапии. Известные данные свидетельствуют о том, что этот процесс активно контролируется на молекулярном уровне и играет одну из определяющих ролей в регуляции и функциональной активности антител. Однако механизмы, лежащие в основе этого контроля, до конца не понятны. Будущие исследования должны быть направлены на изучение передачи сигналов во время продукции иммуноглобулинов В-клетками, что позволит разгадать механизмы, лежащие в основе специфического гликозилирования антител. Это позволит установить ключевые пути реализации эффек- торной активности антител. Полученные таким образом данные дадут новый толчок в разработке терапевтических препаратов и вакцин следующего поколения. Всё это откроет новую главу в терапии широкого спектра инфекционных, онкологических и аутоиммунных заболеваний.
