Заключение
За последние годы широкое использование современных методов молекулярной генетики и протеомики позволило значительно уточнить и конкретизировать наши представления о факторах вирулентности патогенных стрептококков. С помощью биоинформационного анализа полного генома у патогенных штаммов S. pyogenes были идентифицированы гены, ответственные, в частности, за способность противодействовать защитным функциям макрофагов: мобилизации в очаг инфекции, захвату бактерий, их внутриклеточной гибели. Названы конкретные гены, контролирующие отдельные механизмы агрессии, значительная часть которых направлена на подавление защитных функций макрофагов. Кроме того, выявлена сложная сеть генов глобальных транскрипционных регуляторов экспрессии многих факторов вирулентности S. pyogenes.
Показано, что на разных стадиях инфекции и взаимодействия с макрофагами геном S. pyogenes претерпевает значительные изменения в направлении повышения экспрессии факторов вирулентности. Экспериментально доказанная способность S. pyogenes выживать и пролиферировать внутри клеток организма-хозяина, в том числе в макрофагах, заставляет пересмотреть классическое представление о принадлежности S. pyogenes к внеклеточно паразитирующим бактериям. Уточнение природы факторов вирулентности S. pyogenes может стать основой поиска средств их нейтрализации в качестве одного из путей лечения инфекции, вызванной инвазивным патогеном. Детальная оценка путей регуляции взаимодействия макрофагов с S. pyogenes может быть положена в основу разработки новых лекарственных средств, направленных на активацию механизмов врожденного иммунитета против S. pyogenes.
